3-4 COBE 2991 Cell Processor Essentials Guide
Operator Controls
It is not possible to advance directly from Start/Spin to Agitate/Wash-In. To bypass
the Supernatant-Out operation, you must press the SUPER OUT button and then
immediately press the AGITATE/WASH-IN button.
AGITATE WASH IN
When you press the AGITATE WASH IN button, the COBE 2991 advances to the
Agitate/Wash-In operation and the AGITATE WASH IN light illuminates. During
the Agitate/Wash-In operation, the hydraulic pump reverses its direction, and the
centrifuge decelerates. When the hydraulic pump reaches its home (starting)
position, the selected or programmed valve (V1, V2, or V3) opens (see
“VALVE
SELECTOR” on page 3-3) and the centrifuge starts agitating (oscillating). The
AGITATE WASH IN button is only active during a Supernatant-Out operation.
In Automatic mode, the AGITATE WASH IN light illuminates when the COBE
2991 advances to the Agitate/Wash-In operation from the Supernatant-Out
operation.
In both Automatic and Manual modes, pressing the AGITATE WASH IN button
during a Supernatant-Out operation immediately advances the COBE 2991 to the
Agitate/Wash-In operation.
If no valve is programmed to open, a Hold status occurs at the beginning of the
Agitate/Wash-In operation. For information on programming the “no valves
option, see
“VALVE SELECT” on page 4-5 for COBE 2991s with an LED
program board or “VALVE” on page 5-4 for COBE 2991s with a pin-diode
program board.
PREDILUTE
When you press the PREDILUTE button, valves V1 and V2 open, allowing
processing/wash solution to flow into either the cell processing bag or the cell
product bag.
To predilute the starting cell product, the clear tubing below the manifold junction
must be clamped. This prevents processing/wash solution from entering the cell
processing bag.
READY
The READY status light indicates that the COBE 2991 is in the Ready status. For
the READY light to illuminate, the power must be turned on, the hydraulic pump
must be at its home (starting) position, the sliding centrifuge covers must be closed,
and the centrifuge cover interlock latch must be closed (if applicable). The
centrifuge cover interlock latch is present only on newer COBE 2991s; see
“Centrifuge Cover Interlock Latch” on page 1-8.
The COBE 2991 must be in the Ready status before processing can begin.
