Alternatively, it is possible to connect the starting cell product to the blue-striped
tubing and use the red-striped tubing (valve V1) for additional processing/wash
solution (
Figure 6-14). The Blood-In and Air-Out operations cannot be used if the
starting cell product is connected in this manner. See “Loading Cell Product Using
the Blue-Striped Tubing” on page 6-23 for more information.

Predilution and Loading
Predilution reduces the cell concentration to ensure adequate washing. It also
facilitates the flow of starting cell product in and out of the cell processing bag.
To ensure adequate washing, the maximum volume of red blood cells (RBCs)
loaded into the cell processing bag should not exceed 450 mL.
From the following list, select the appropriate procedure to predilute the starting
cell product (if necessary) and load the cell processing bag.
• If the starting cell product has a hematocrit of less than 35%, proceed to
“Loading Cell Product with no Predilution (Hematocrit of 35% or Less)” on
page 6-16.
• If the starting cell product has a hematocrit greater than 35%, and it will be
prediluted with processing/wash solution from the green-striped tubing,
proceed to
“Predilution Procedure Using the Green-Striped Tubing
(Hematocrit of 35% or Greater)” on page 6-18.
• If the starting cell product has a hematocrit greater than 35%, and it will be
prediluted with processing/wash solution from the blue-striped tubing,
proceed to
“Predilution Procedure Using the Blue-Striped Tubing” on
page 6-21.
• If the starting cell product is connected to the blue-striped tubing, proceed to
“Loading Cell Product Using the Blue-Striped Tubing” on page 6-23.
