Processing and Common Operations
Loading the Starting Cell Product
The following sections provide a generic overview of connecting the starting cell
product and solutions, predilution, and loading. For instructions regarding specific
protocols, refer to the COBE 2991 Cell Processor Protocols Guide.
Connecting the Starting Cell Product and Solutions
The starting cell product is typically connected to the red-striped tubing (through
valve V1), with processing/wash solutions connected to the green and yellow-
striped tubing (through valves V2 and V3, respectively). This configuration
(Figure 6-13) allows the use of the Blood-In and Air-Out operations to load the
starting cell product and remove trapped air from the cell processing bag. You have
the option of connecting additional processing/wash solution or a collection
container to the blue-striped tubing.

If you are using the red-striped tubing to load the starting cell product, use the
following procedure:
1 Remove the cap from the spike on the green-striped tubing, and insert the
spike into a processing/wash solution container. Hang the container on the
right hanger bar.
2 If additional processing/wash solution is required, remove the cap from the
spike on the yellow-striped tubing, and insert the spike into a processing/wash
solution container. Hang the container on the right hanger bar.
3 Remove the cap from the spike on the red-striped tubing, and insert the spike
into the starting cell product container. Hang the container on the left hanger
bar.
4 Close the slide clamp on the blue-striped tubing.
If you are using the blue-striped tubing, remove the cap from the spike on the
blue-striped tubing and insert the spike into a collection container or
processing/wash solution container. Hang additional processing/wash solution
on the left hanger bar.
Caution: Use
aseptic technique
while connecting
cell product and
processing/wash solutions
to the cell processing set.
