System Description
COBE 2991 Cell Processor Essentials Guide 1-9
Introduction
1
Hydraulic System
The hydraulic system (shown in Figure 1-2 on page 1-4, refer to items r through u)
consists of a piston-type pump assembly, flow controls and switches, a network of
piping, and a hydraulic fluid reservoir. Hydraulic fluid is pumped underneath the
flexible diaphragm to express cells or supernatant out of the cell processing bag.
Hydraulic Fluid
The fluid used in the hydraulic system is a mixture of ethylene glycol and water. It
has a specific gravity of approximately 1.07 ±.01. This particular specific gravity is
used to enhance the separation of white blood cells (WBCs) and plasma from
RBCs. To facilitate the separation of other types of cells, adjustments may be made
to the specific gravity of the hydraulic fluid.

The cell processing set (Figure 1-8 and Figure 1-9 on page 1-10) is sterilized with
ethylene oxide and is latex-free. It consists of a cell processing bag, five color-coded
tubing segments connected by a junction manifold, a pre-attached waste collection
bag, and a rotating seal. Four tubing segments with pre-attached spikes allow
starting cell product and processing/wash solutions to be added or removed from
the cell processing bag. Waste can be diverted to the pre-attached waste collection
bag. Expressed fluids or cells can be diverted to an optional product collection
container. The rotating seal allows the cell processing bag to rotate while the
centrifuge is spinning and provides a pathway for fluid to be removed and/or added
during processing. The cell processing bag has a capacity of approximately 630 mL
when placed in the centrifuge bowl and has a spike receptor for easy removal or
transfer of the ending cell product.
The triple processing set (Figure 1-9), commonly used for bone marrow processing,
has three cell processing bags, additional connectors, and a special tubing segment
for use with a peristaltic pump.
A double coupler adapter (Figure 1-10) and female luer connector (Figure 1-11) are
also available, giving you additional connection options.
